The Gospel Oak to Barking Overground line will be closed on Sunday.
Network Rail said essential work will be completed on the route between 12.30am on Sunday and 3.45am on Monday.
The delivery of materials and other less disruptive work will take place outside of those hours to make full use of the time available.
Residents near the line have been warned to expect some noise caused by vehicle movements.
Engineers have been working to raise the height of the parapets on Queens Road Bridge to 1.8m to comply with safety standards.
Replacement buses will operate between Gospel Oak and Seven Sisters.
